Each 0.1 ml dose contains: Purified protein derivative of bovine tuberculin from culture of Mycobacterium bovis, strain AN5. 3000 IU/dose. Excipients q.s.
In all species apply intradermally: 0.1 ml of Bovine Tuberculin PPD 3000 containing 3000 IU/dose (≥ 66% and ≤ 150%). Bovine Tuberculin PPD 3000 is a diagnostic reagent for in vivo identification of the disease in cattle, sheep, goats and swine that have been exposed to tuberculosis-causing Mycobacterium species. Age of application of Bovine Tuberculin PPD 3000 depending on the animal species and the type of farm they are destined to (according to SENASA128/2012 resolution): Cattle: dairy farms, mil dairy or beef cattle farms. Artificial insemination or embryo transfer centers: apply in cattle from 3 months of age. Meat: apply in cattle from 12 months of age. Goats: Milk and meat production: apply in animals from 3 months of age. Sheep: Dairy farms: apply in sheep from 6 months of age. Swine: Apply to the whole herd.
Contraindications: None. The doses, application sites and test readings in different animal species are described below.
Cattle
Selection of injection sites. The injection sites for bovine species are in the caudal fold, simple or comparative cervical fold. In animals that have inflammation or unrelated lumps, close to the recommended injection site, tuberculin should be injected on the opposite side of the animal. Caudal site: Intradermal injection in the middle third of the internal caudal fold 6 cm from the base of the tail. Tuberculin test with bovine PPD in the caudal fold is a routine procedure. Cervical site (single injection): The injection site is located on one side of the neck, in the center of the middle third. The single cervical test with bovine PPD is for sanitation. Cervical site (double injection: comparative test): Approved injection sites are located at the boundary between the anterior third and middle third on either side of the neck. To establish whether cattle are infected with Mycobacterium bovis or other non-bovine strains of mycobacteria, the comparative test is performed by simultaneously inoculating the neck (neck’s injection triangle) with bovine PPD and avian PPD. Results (reaction) are expressed as the difference in skin thickness (mm) between the pre- and post-skin test readings. Avian PPD was chosen as a term of comparison, in principle, to rule out Mycobacterium avium infection and also because this bacterium has common antigenic relationships with the species Mycobacterium avium subsp. paratuberculosis (Paratuberculosis) and with others responsible for cross-reactions to Bovine Tuberculin. In adult animals the highest inoculation site is approximately 10 cm below the nuchal ligament for avian PPD. The lowest site should be approximately 12 cm from the upper site where bovine PPD will be inoculated, on a line parallel to the shoulder line. In young animals where there is insufficient space to inject both types of PPD tuberculin on the same side of the neck, they should be injected in the center of the middle third on each side of the neck, respectively. In animals with unrelated inflammation or lumps close to the injection site(s), PPD tuberculin should be injected on the opposite side of the neck.
Sheep
They are also susceptible to infection with M. avium subsp paratuberculosis, which causes paratuberculosis disease, therefore the comparative test with bovine PPD and avian PPD tuberculin should be used. 0.1 ml of bovine tuberculin is injected intradermally into the skin of the right armpit and 0.1 ml of avian tuberculin is injected intradermally into the skin of the left armpit, with two (2) different syringes. The reading should be performed 72 hours after administration by palpation and measured with a caliper.
Swine
Both bovine PPD and avian PPD tuberculin should be used. Inject (0.1 ml) of bovine tuberculin and (0.1 ml) of avian tuberculin, with two (2) different syringes. The intradermal injection site is the skin at the base of the ear. If this region is scarred or otherwise defective, testing can be done on the lips of the vulva of the sow or at the junction between the male’s rectal mucosa and the skin. Bovine tuberculin is injected on the right side and avian tuberculin on the left. The reading is performed after 48 hours, and any tissue reaction verified by palpation is classified as reactive, and it should be noted whether it is due to bovine or avian tuberculin.
Goats
It should be applied in the caudal fold or on the neck. The technique of application, reading and interpretation is similar to that of cattle.
